Robert Plant once said something along the lines of "Stairway to Heaven" being the prototypal rock song, but that "Kashmir" was the prototypal Led Zeppelin song.
I think it depends on what you mean by "greatest". You can go by chart action (its peak position, how long it was on the charts), or (my preference) the songs that you instantly identify with an artist. Using the second definition, here are the songs I'd pick...
Rolling Stones - Satisfaction
Beatles - Hey Jude
Pink Floyd - Another Brick In The Wall (yeah, I know, easy pick)
Michael Jackson - Billie Jean
Devo - Whip It
The Knack - My Sharona. That's the only Knack song most people know.
The Who - Pinball Wizard
Rolling Stones? Satisfaction
Beatles? Paperback Writer
Pink Floyd? Money
Michael Jackson? Beat It
ZZ Top? the one about the sunglasses
Devo ? Whip It
The Knack? My Sharonna
The Who? Won't Get Fooled Again
Of course there are objective answers possible, e.g. total sales or weeks on the chart.
Aerosmith - Dream On
Chicago - 25 or 6 to 4
Blood, Sweat & Tears - Spinning Wheel
Cream - Sunshine of Your Love
Iron Butterfly - Inna-Gadda-Da-Vida
Derek & the Dominoes - Layla
Brian Wilson - Good Vibrations
Paul McCartney (with Beatles) - Hey Jude
Paul McCartney (solo) - Band on the Run
John Lennon (with Beatles) - Come Together
John Lennon (solo) - Give Peace a Chance
George Harrison (with Beatles) - Something
George Harrison (solo) - All Things Must Pass
Ringo Starr (with Beatles) - Yellow Submarine / Octopus's Garden (tie)
Ringo Starr (solo) - I Am the Greatest
Rolling Stones? - My personal choice is Sympathy for the Devil.Satisfaction is one hell of a song, but I really think Sympathy is the one that keeps the Stones from being just another pop band. Beatles? - This is an almost impossible pick, as their sound changed so much depending on the era. I guess I'll go with I Saw Her Standing There. Pink Floyd? - I'm going to go with Time Michael Jackson? - Billie Jean ZZ Top? - La Grange. As tempting as it is to pick one of their 80's hits, I think this is more iconic. Devo - Whip It is the easiest choice, as I'm pretty sure it was the only Devo song to break the top 40. However, I think I would choose Two Tracks instead, as it's just more of a Devo song. The Knack? - Well, the obvious choice is My Sharona--so I'll pick Good Girls Don't instead, if for no other reason than to be conrary.. The Who - Either My Generation or The Real Me

It seems there haven't been many suggestions for ZZ Top. I think Legs is there signature song.
Queen -Bohemian Rhapsody or We Will Rock You/We Are The Champions?.
