I'd like to see if I can get this list up to one thousand. I'm sure that I've missed a lot of obvious ones, and even more obscure ones.

There's Electra Woman and Dyna Girl from the '70s kid-vid show of the same name.
G-Girl from the film My Super Ex-Girlfriend.
Darla Dudley, who shares the power of Shazam in Shazam!
Queen Atlanna from Aquaman.
Jetstream, Royal Pain, Principal Powers, Freeze Girl and Nurse Spex (and maybe others) from Sky High.
Catgirl (Julie Carroll)
-UK. Comic Book Plus link.
The Fabuous Four: Hercula, Helen, Electra and Fauna
-UK. From annuals, but they appeared in several stories. The stories were drawn by Enrique Romero.
The various heroines from the two runs of the British TV show The Tomorrow People:
-First run: Carol (series one); Elizabeth (from season two); Hsai Tai (from season six). I don't think Patricia qualifies. She was more an antagonist who became an ally of the heroes.
-Second run: Lisa ("The Origin Story"); Ami (from "The Culex Experiment"); Jade (as a character with superpowers, "The Living Stones" - she develops them near the end). I can't remember if Lisa does anything to help others in her story or is only menaced. Off the topic, Christopher Lee played the villain in "The Rameses Connection".
-There was an American version of the show more recently, but I know nothing about it.
Sharron Macready from another British show, The Champions.
Kate Mason, the Sandra Bullock character from Bionic Showdown.
Gloria Dinallo, the Courteney Cox character from Misfits of Science.
I couldn't remember this show's name, and trying to find it I learned of the TV show No Ordinary Family, which I've never seen. Some of its characters might qualify.
No thoughts on these?

There was Fleeta, the partner of Dell's Dracula who could also turn into a bat.
And there was another Batwoman who was an alien crimefighter who resembled a vampiress from Super Friends #10. Her like-powered husband was known as...Batman!
Leeta 87 was a super-heroine in the future who died when she slipped on an alien banana peel as seen in Adventure Comics #341 (F'66).
Then there's Lamprey and Nightwind, two Legion applicants who first appeared in Legion of Super-Heroes #272.
Let's go over everyone's suggestions (plus a few of my own) at once:
Notes: Heard of The Tomorrow People (mostly in Doctor Who DVD extras, but never seen any of it. Never eveve knew there was an American verssion.
The Fabulous Four seem to have been also known as the Supercaen.
Sorry, I missed those, somehow. You guys are faster than I am.

OK, wow. Unlike some of my other lists, this one was a real cooperative effort. It would not have grown the way it did without all of the following (with profound apologies to anyonewhose name I missed):
Without your time and effort, this list ould be nowhere near one thousand, let alone beyond it.
One great thing about this list is that I have leanred a lot about so many characters that I never even suspected existed.
DC's Enchantress (June Moon) started out as a super-heroine in the Silver Age with a brief run in her own strip in one of their mystery titles. When she finally reappeared in the Bronze Age in Superman Family, an attempt to increase her powers went awry, putting her into conflict with Supergirl. Eventually, she was scooped up into the "Forgotten Villains" group (rather than getting a redemption arc to put her in the "Forgotten Heroes" team), which led to her joining the Suicide Squad, the only reason she wasn't actually forgotten.

Happy to help. I marvel at you having created these other lists without our assistance. Plus, I shared Captain Comics' embarrassment at the prospect that we couldn't come up with a thousand worthy candidates.
